Function | Component of the type IV secretion system Dot/Icm that is required for intracellular growth in human macrophages and amoebae. Probably Dot/Icm system acts during phagocytosis to prevent phagosome-lysosome fusion and to establish the L.pneumophila replication vacuole, a distinct niche that allows intracellular replication of the bacterium.
The Icm/Dot system may also be involved in conjugation. Deletion experiments show that DotB protein can be sustituted by homologous component of the lvh type IV secretion for L.pneumophila function during conjugation, but not for intracellular growth. Together with virB4, may be implicated in providing the energy, via hydrolysis of ATP, for the assembly of secretion system and substrate transport.
