2ndcategory | Uncharacterized Protein Family IcmO/DotL |
3rdcategory | Effector translocator |
Function | Component of the type IV secretion system Dot/Icm that is required for intracellular growth in human macrophages and amoebae. The IcmO/DotL protein is not only required for growth of L. pneumophila inside macrophages but is also essential for viability of certain strains on bacteriological media.

Miscellaneus | DotL is an inner membrane protein. A model proposed by Buscher et al (2005) says that DotL play the role of a regulator of the T4SS, controlling the opening and closing of the pore. DotL contains a nucleotide binding motif and shows extensive sequence similarity to the conjugal transfer protein TrbC from IncI plasmids. Also has limited homology to a family of proposed ATPases known type IV secretion system coupling proteins (T4CPs).
