Function Component of the type IV secretion system Dot/Icm that is required for intracellular growth in human macrophages and amoebae. The IcmO/DotL protein is not only required for growth of L. pneumophila inside macrophages but is also essential for viability of certain strains on bacteriological media.
Miscellaneus DotL is an inner membrane protein. A model proposed by Buscher et al (2005) says that DotL play the role of a regulator of the T4SS, controlling the opening and closing of the pore. DotL contains a nucleotide binding motif and shows extensive sequence similarity to the conjugal transfer protein TrbC from IncI plasmids. Also has limited homology to a family of proposed ATPases known type IV secretion system coupling proteins (T4CPs).
